Isoflavone Content and Estrogen Activity in Arrowroot Puerariae Radix

Abstract
The content of isoflavone and the estrogenic activity in Puerariae Radix were investigated. HPLC using a dual column system was used to determine the content of isoflavone in Puerariae Radix. The simultaneous analytical time of isoflavones (daidzin, genistin, daidzein, glycitein, genistein) required less than 25 min per sample. The content of isoflavones (daidzin, genistin, daidzein, glycitein, genistein) required less than 25 min per sample. The content of daidzin (5,026-18, 547 ¥ìg/g) genistin (488-1,201 ¥ìg/g), daidzen (316-876 ¥ìg/g), genistein (8-63 ¥ìg/g), puerarin (1-10 g/100 g) in Pueraria thunbergiana showed higher levels compared to content of daidzin (168 ¥ìg/g) genistin (62 ¥ìg/g), daidzein (46 ¥ìg/g), puerarin (0.03 g/100 g) in Pueraria mirifica. Glycitein was not detected in Puerariae Radix. In uterotrophic assay as indicator of estrogenic activity, the Pueraria mirifica exhibited strong estrogenic activity, whereas Pueraria thunbergiana and Butea superba did not. Therefore, the content of isoflavones in Puerariae Radix could no be responsible for estrogenic activity.
